A Study on the Cellular Fatty Acid Profiles of V. parahaemolyticus Using Gas Chromatography

Abstract
This study was carried out to isolate Vibrio parahaemolyticus in Pusan areas and to analyze whole cell fatty acid methyl ester profiles using gas chromatography with an application of the flexible, fused silica capillary colume for increased resolution.
The results of the study were as follows
1. The biochemical properties were typical characteristics to V. parahaemolyticus and 23 strains(77 %) were positive reaction in the L-arabinose + 1 % NaCl.
2. The results of serotypes of the isolated V. parahaemolyticus were composed of 8 strains (27%) on K I group, 4 strains(13 %) on K II group, 2 strains(? %) on K I1 group, 5 strains (17 %) on K IV group, 5 strains (17 %) on K V group, 2 strains (7 %) on K VII group, 4 strains (13 %) on K VIII group.
3. The cellular fatty acids of V. parahaemolyticus were C12:0, C12:0 30H, C14:0, C15:0, C16:1 CIS 9, C16:1 C, C16:0, C17:0 ISO, C17:1 B, C17:0, C18:0, summed feature 3 (C16: 1 ISO 1 /14:0, 30H or C14:0 30H/16:1 ISO I ), summed feature 7 (C18:1 CIS 11/t 9/t6 or C18:1 TRANS 9/t6 /c 11 or 18:1 TRANS 6/9/cll) and 3.08%,1.87%, 9.08%,1.24%, 37.46 %, 0.31 %, 23.75 %, 0.32 %, 0.82 %, 0.87 %, 0.66 %, 2.72 %, 16.38 % respectively.
4. C10:0 30H, C16:1 ISO E, C16:0 ISO, C16:1 B, C18:1 CIS 9 were found in V. cholerae non 01, but those were not found in V. prahaemolyticus. C12:0, C15:0, C16:1 C, C17:0, ISO, C17:0 were observed in V. parahaemolyticus, those were not in V. cholerae non 01. C12:0, C15:0, C16:0 ISO, C16:1, C17:0 were observed only in V. vulnificus.
This study, in conclusion, demonstrated a method to isolate and to identify V. parahaemolyticus rapidly through analyzing fatty acids using gas chromatography, and it demonstrated that it was possible to identify bacterial species through comparing the composition of fatty acids of Genus Vibrio.
